ABSTRACT

The chemical constituents of classical prescription Danggui Buxue Decoction were analyzed by reversed-phase(RP) chromatography and hydrophilic interaction chromatography(HILIC) coupled with quadrupole time-of-flight mass spectrometry. RP separation of Danggui Buxue Decoction was performed on ACQUITY UPLC HSS T3(2.1 mm×100 mm, 1.8 μm), while HILIC separation was on Waters BEH Amide(2.1 mm×100 mm, 1.7 μm). Mass spectrometry(MS) data were acquired in both negative and positive ion modes. Chemical constituents of Astragali Radix and Angelicae Sinensis Radix were searched from Reaxys and thus the in-house library was established. MS data were further analyzed by MassLynx 4.1 combined with in-house library, HMDB, Reaxys, and comparison with reference substances. In conclusion, a total of 154 compounds were identified and characterized: 16 saponins, 44 flavonoids, 10 phthalides, 7 phenylpropanoids, 15 bases and the corresponding nucleosides, 30 oligosaccharides, and 32 other compounds. Among them, 65 compounds were detected by HILIC-MS/MS. This study provides experimental evidences for the material basis research, quality control, and preparation development of Danggui Buxue Decoction and a reference method for comprehensive characterization of Chinese medicine decoctions typified by classical prescriptions.

ABSTRACT

To clarify the key quality attributes of substance benchmarks in Danggui Buxue Decoction(DBD), this study prepared 21 batches of DBD substance benchmarks, and established two methods for detecting their fingerprints, followed by the identification of peak attribution and similarity range as well as the determination of extract and transfer rate ranges and contents of index components ferulic acid, calycosin-7-O-β-D-glucoside, and astragaloside Ⅳ. The mass fractions and transfer rates of DBD substance benchmarks from different batches were calculated as follows: ferulic acid(index component in Angelicae Sinensis Radix): 0.037%-0.084% and 31.41%-98.88%; astragaloside Ⅳ(index component in Astragali Radix): 0.021%-0.059% and 32.18%-118.57%; calycosin-7-O-β-D-glucoside: 0.002%-0.023% and 11.51%-45.65%, with the extract rate being 18.4%-36.1%. The similarity of fingerprints among 21 batches of DBD substance benchmarks was all higher than 0.9. The quality control method for DBD substance benchmarks was preliminarily established based on the HPLC fingerprint analysis and index component determination, which has provided a basis for the subsequent development of DBD and the quality control of novel related preparations.

ABSTRACT

Danggui Buxue Decoction is a classic prescription of Qi and blood tonification, which is mainly applied in treating fatigue, internal damage Qi weakness, blood deficiency, and outward going of floating Yang. Modern pharmacology shows that it can promote hematopoiesis, regulate immunity, and protect heart and cerebral vessels. The prescription, often used for the treatment of anemia and other diseases in clinic, is composed of Astragali Radix and Angelicae Sinensis Radix at a dosage ratio of 5∶1. It is a modern compound prescription for invigorating Qi and generating blood. Based on the review of the chemical constituents, pharmacological effects, and clinical applications of Danggui Buxue Decoction, its Q-marker was predicted and analyzed according to the "five principles" of Chinese medicine Q-marker--quality transmissibility and traceability, ingredient specificity, component validity, component measurabi-lity, and formula compatibility environment. The results suggested that calycosin, calycosin-7-O-β-D-glucoside, formononetin, ononin, astragaloside A, ferulic acid, and ligustilide could be used as Q-markers of Danggui Buxue Decoction, which provides reference for establishing the quality system of Danggui Buxue Decoction.

ABSTRACT

Colon cancer-related anemia (CCRA) is mainly caused by systemic inflammation, intestinal bleeding, iron deficiency and chemotherapy-induced myelosuppression in colon cancer. However, the best therapeutic schedule and related mechanism on CCRA were still uncertain. Studies on blood enrichment and anti-tumor effects of combined Danggui Buxue Decoction (DBD), Fe and rhEPO based on CCRA and gut microbiota modulation were conducted in this paper. Here, CCRA model was successfully induced by subcutaneous inoculation of CT-26 and i.p. oxaliplatin, rhEPO + DBD high dosage + Fe (EDF) and rhEPO + DBD high dosage (ED) groups had the best blood enrichment effect. Attractively, EDF group also showed antitumor activity. The sequencing results of gut microbiota showed that compared to P group, the relative abundances of Lachnospiraceae and opportunistic pathogen (Odoribacter) in ED and EDF groups were decreased. Interestingly, EDF also decreased the relative abundances of cancer-related bacteria (Helicobacter, Lactococcus, Alloprevotella) and imbalance-inducing bacteria (Escherichia-Shigella and Parabacteroides) and increased the relative abundances of butyrate-producing bacteria (Ruminococcaceae_UCG-014), however, ED showed the opposite effects to EDF, this might be the reason of the smaller tumor volume in EDF group. Our findings proposed the best treatment combination of DBD, rhEPO and Fe in CCRA and provided theoretical basis and literature reference for CCRA-induced intestinal flora disorder and the regulatory mechanism of EDF.

ABSTRACT

Danggui Buxue Decoction is recorded in The Theories of Differentiation on Endogenous by Dong-yuan Li in Jin Dynasty. It consists of Astragali Radix and Angelicae Sinensis Radix with a weight ratio of 51, which is a classic Chinese medicinal prescription for benefiting qi and nourishing blood. Clinically, it is mainly used in treating anemia, regulating immune function, protecting cardiovascular and cerebrovascular system, anti-tumor and so on. This paper summarizes and analyzes the research on the material basis of Danggui Buxue Decoction by referring to the Chinese and foreign literatures, so as to provide reference for the further research and application of Danggui Buxue Decoction.

ABSTRACT

Diabetes mellitus (DM) is a chronic metabolic disease characterized by hyperglycemia. Its main complications of diabetes, such as diabetic angiopathy, have seriously affected the quality of life for patients, and have become an important cause of death and disability. The underlying pathological changes include macrovascular lesions and microvascular lesions. Diabetic macrovascular lesions mainly involve thoracic aorta, coronary artery, carotid artery, cerebral artery and peripheral blood vessels, etc., and the common clinical diseases include coronary heart disease, stroke, peripheral neuropathy, lower extremity arteriosclerosis, etc. Diabetic microvascular lesions mainly involve the heart, brain, kidney and other microvessels. Nowadays, various new oral hypoglycemic agents and insulin have emerged in the society and are widely used in clinical practice. However, traditional Chinese medicines(TCMs) have stable curative effect, less side effect, and can improve glucose metabolism, lipid metabolism, insulin resistance, oxidative stress, expression of inflammatory cytokines, vascular endothelial injury, microcirculation disorders, balance of fibrinolysis system and blood coagulation system, and improve the syndromes of TCMs, etc. They have been widely recognized and applied in the prevention and treatment of diabetic angiopathy. A profound understanding on the etiology, pathogenesis and treatment of diabetic angiopathy has been formed in Chinese medicine. Therefore, in this paper, we would summarizes the understanding on Chinese medicine for diabetic angiopathy and the mechanism of Yiqi Huoxue prescription in the treatment of diabetic angiopathy in the past three years.

ABSTRACT

Objective: Taking Danggui Buxue Decoction (DBD) as the research object, the differences of chemical composition types and content among traditional decoction, self-made granule decoction and marketable granule decoction were compared, in order to provide scientific basis for clinical application of Chinese medicine formul granules. Methods: The fingerprint was established by HPLC. The value of the chemical composition, the content of the index components, the area of the common peak area, and the similarity of the fingerprint were evaluated, and the different dosage forms of the decoction of DBD were investigated. The effect of chemical composition on the chemical equivalence of traditional decoction and formula granules was compared. Results: The number of chromatographic peaks ranged from 15 peaks of traditional decoction to 13 (factory A), 12 (self-made), 11 (factory B), and 9 (factory C). The content of ferulic acid in formula granules was significantly different from that in traditional decoction (P formula granule decoction of factory A > self-made formula granule decoction > formula granule decoction of factory B > formula granule decoction of factory C. Content of campanulin was in order : self-made formula granule decoction > traditional decoction > formula granule decoction of factory A > formula granule decoction of factory B > formula granule decoction of factory C. The total peak area of formula granule decoction was lower than that of traditional decoction. If the traditional decoction was 1, the others (self-made and factories A, B, C) were equivalent to 0.95, 0.66, 0.40 and 0.47, respectively. Comparing with traditional decoction, self-made formula granule decoction and formula granules from factories A and B had higher similarity (0.97, 0.96, 0.98), while that from factory C was slightly lower (0.85). The main chromatographic peaks of DBD were found to be from the single herbs and no new chemical components were found. The disappearance of ingredients in formula granule Decoction was found. Conclusion: The content of index components and the number of chromatographic peaks of traditional decoction of DBD is higher than those in the formula granule decoction. There are some differences between them. This indicates that the clinical equivalence of formula granules is not consistent with the reality of decoction, so the recommended clinical equivalence of formula granules of traditional Chinese medicine should be corrected to promote rational clinical application, which can provide scientific research ideas for the unified management of the quality of formula granules of Chinese materia medica.

ABSTRACT

To explore the pharmacodynamic material basis and mechanism of the classic TCM excellent prescriptions(cTCMep) Danggui Buxue decoction, so as to provide more choices for the prevention and treatment of diabetes mellitus (DM). Methods With the integrative pharmacology of traditional Chinese medicine (TCMIP) , we predicted the pharmacodynamic material basis and mechanisms of the cTCMep Danggui Buxue decoction for treatment of DM. Results The 50 " Astragalus-Angelica" components in the cTCMep Danggui Buxue decoction directly or indirectly acted on 60 key drug targets. These drug targets restored the disease imbalance network of DM and its complications by acting on 30 major regulatory pathways such as nervous-endocrine system, gap junction, hormone signaling pathways, and cardiovascular circulatory system. The main components of Astragalus membranaceus membranaceus involved in the prevention and treatment of DM were astragalosides, astragalosides and alavonoids. The main components involved in the prevention and treatment of DM in Angelica sinensis were organic acids such as ferulic acid, oil components such as (3-sweet myrrh, stigmasterol-|3-D-glucoside, smbelliferol and scopolamine. Conclusions The 50 " Astragalus-Angelica" components in the cTCMep Danggui Buxue decoction may play a role in preventing and treating DM through the AVPR1A, AVPR1B and AVPR2 (AVPR: Arginine vasopressin receptor, three subtypes), of which Astragalus membranaceu is the key component.

ABSTRACT

Objective To optimize extraction technology of Compound Danggui Buxue Decoction. Methods On the basis of single factor test, factorial design, and fastest rise test, yield and weight sum of extraction efficiency of ferulic acid, paeoniflorin and ligustilide were set as response, and central composite design-response surface method was used to optimize the extraction technology of Danggui Buxue Decoction. Results The optimum conditions consisted of ultrasonic-wave extraction for 60 min, ethyl alcohol concentration 36% and the ratio of material to solvent = 1:94. Conclusion This method has high extraction efficiency, easy operation and good predictability.

At present, the drugs to treat DD are mainly chemical drugs.Due to the single target, strong drug dependence and obvious side effects, etc., it is a tough problem to be solved urgently in clinical practice to find a treatment drug with multiple targets, low toxicity and little side effects. The research group found that the invigorating qi and nourishing blood formula Danggui-BuxueDecoction (DBD) has a relatively exact effect on DD.

AIM To study the effects of Danggui Buxue Decoction total glycosides combined with Hirudo in the treatment of pulmonary fibrosis in rats and its mechanism.METHODS One hundred and twenty male SD rats were selected,eighteen of which were taken as normal group (normal saline),the remaining 102 rats were used to establish model for pulmonary fibrosis.Seventy-two modeled rats were randomly divided into model group (normal saline),Danggui Buxue Decoction total glycosides group (13 mg/kg),Hirudo group (Hirudo powder,4 g/kg) and combination group (Danggui Buxue Decoction total glycosides,13 mg/kg;Hirudo powder,4 g/kg),eighteen rats in each group with intragastric administration.RESULTS On the 7th,14th and 28th days after modeling,alveolar inflammatory score,pulmonary fibrosis degree score,TGF-β1 and PAI-1 expression levels in lung tissue,IL-4 and IL-17 expression levels in alveolar lavage fluid,hydroxyproline (HYP) content in lung tissue in the model group were significantly higher than those in the normal group (P < 0.05).Alveolar inflammatory score,pulmonary fibrosis degree score,TGF-β1,PAI-1,IL-4,IL-17 expression levels and HYP content in the Danggui Buxue Decoction total glycosides group,Hirudo group and combination group were significantly lower than those in the model group (P < 0.05).In the combination group,alveolar inflammatory score,pulmonary fibrosis degree score,TGF-β1,PAI-1,IL-4,IL-17 expression levels and HYP content were significantly lower than those in the Danggui Buxue Decoction total glycosides group and Hirudo group (P < 0.05).CONCLUSION Danggui Buxue Decoction total glycosides combined with Hirudo have good therapeutic effects on pulmonary fibrosis in rats mainly by reducing TGF-β1,PAI-1 expression levels and HYP coment in lung tissue.

ABSTRACT

Objective To evaluate the effect ofJiawei-Danggui-Buxue decoction in the treatment for the patients with hemorrhagic anemia after total knee arthroplasty (TKA).Methods A total of 63 TKA patients with hemorrhagic anemia were included, and were divided into the treatment group (31 patients) and the control group (32 patients). All patients started to take medicine the second day after surgery. The control group took the Ferrous Succinate Tablets and the treatment group took Jiawei-Danggui-Buxue decoction in the following 4 weeks. The Hb and Hct were detected,and the changes of TCM syndrome scores and the total effect rates of anemiawere observed.Results The total effect rate of TCM syndrome scores in the treatment group was 93.54%, which was significantly higher than control group 83.87% (P<0.05); The total effect rate of anemia in the treatment group was 90.32%, which is significantly higher than control group 75.00% (P<0.05). At the fifteenth day after operation,. And at the postoperative 29th day, the levels of Hemoglobin (129.61 ± 13.02 g/Lvs. 113.35 ± 15.46 g/L,t=6.245) and Hematocrit (0.45% ± 0.03 %vs. 0.41% ± 0.04%,t=5.962) in the treatment group were significantly higher than those in the control group (P<0.05).Conclusions TheJiawei-Danggui-Buxue decoctionimprovethe Hb, HCT and the clinical effect rates of anemia and TCM syndromes for the patients with hemorrhagic anemia after TKA.

ABSTRACT

The "black" of Chinese materia medica (CMM) has been criticized all the time. The Maillard reaction exists widely in the course of food processing, which affects the product's color and odor. The suitable conditions of Maillard reaction also exist in the processing of CMM preparation. The Maillard reaction has significant impact to the study of CMM's material basis. So the Maillard reaction and its products in the processing of CMM preparation should be studied. Taking the Danggui Buxue Decoction for instance, the chemical composition of the prescription was consulted by the TCMSP of Northwest A&F University, and then the constituents were classified, the possibility of the Maillard reaction occured in the processing of CMM preparation was analyzed. New ideas on study of material basis of CMM will be put forward.

Objective To study chemical ingredients and angiogenesis effects for different compatibility proportions ofDanggui Buxue Decoction.Methods For chemical components, HPLC was performed to study the ingredients (Ferulic Acid, Calycosin-7-glusosido, Calycosin, Formononetin) inDanggui Buxue Decoction. For angiogenesis effects, transgenosis zebrafish model was incubated with different proportions ofDanggui Buxue Decoction.Results With the raise of Astragali Radix content, the synthetic scores of four ingredients in different compatibility proportions increased. When the Astragali Radix content to Angelicae Sinensis Radix content was 8 to 2 and 5 to 1, the scores were the highest. For zebrafish vascular injury model, vessel recoveries in top three were 5:1, 6:1 and 2:1 (Astragali Radix: Angelicae Sinensis Radix).Conclusion From comprehensive analysis of chemical composition and angiogenesis, the best compatibility ratio of Astragali Radix to Angelicae Sinensis Radix is 5:1.

Objective To study the effect of Danggui Buxue Decoction on red cell immune function in mice with morphine abstinence syndrome. Methods 50 mice were randomly divided into normal group,positive control group, high, medium, low dose group,Hindlimb injection of morphine in morphine dependent mice model was established.Erythrocyte membrane were measured in five groups of mice micro viscosity, erythrocyte deformation index; Determination of red cell immune complex rosette rate using the method (CIC), red cell C3b receptor rosette rate;and to observe the five group of regulator factor enhance rate (RFER) and immune adherence inhibitor factor (RFIR) levels.Results Erythrocyte membrane microviscosity in model group were significantly higher than those in normal group, medium group and low group (P<0.05),red cell deformability index was lower than that of normal group, drug medium group and drug group low (P<0.05).Drug high group and medium group,immune complex rosette rate lower than in model group(P<0.05), C3b receptor rosette rate was significantly higher than that in the model group (P<0.05).Conclusion Low dose group and high dose group Danggui Buxue Decoction raises the red cell immune function in morphine withdrawal mice, improves the withdrawal symptoms, rapids rehabilitation of the body, has good clinical value.

Objective To observe the protective effect of Danggui Buxue Decoction on 60 Co-γray irradiated mice. Methods The experimental mice were randomly divided into 5 groups ( the normal control group , model group , Danggui Buxue Decoction of low , middle and high dose groups ) .The mouse except the normal control group were irradiated by 60Co-γray and established the radiation-damage model.Peripheral blood neutrophil count (NEUT), the number of bone marrow nucleated cells ( BMC ) , the activity of serum superoxide dismutase ( SOD ) and the serum levels of malondialdehyde ( MDA) were measured .Results Radiation injury in mice of NEUT , BMC and SOD were significantly decreased (P <0.01), MDA were increased (P <0.01).Danggui Buxue Decoction can increase the levels of NEUT , BMC and SOD in mice after radiation injury , decrease the content of MDA .And protective effect of high dose group was better (P <0.01).Conclusions Danggui Buxue Decoction has protective effect on radiation injury in mice .And there is a correlation between the protection effect and dose .

Objective To explore the protective effect of Sijunzi decoction and Danggui Buxue decoction on the immunological function in tumor-bearing mice treated by cyclophosphamide.Methods Transplantable carcinomas animal model were constructed by inoculating tumor cell to mice for 24 hours,and randomly divided into five groups.Tumor-bearing control group were reveived normal saline 0.2 mL per mice one day by intraperitoneal injection,and normal saline 0.4 mL per mice each day by intragastric administration.Cyclophosphamide group were reveived CTX 20 mg per kg weight each day,and normal saline 0.4 mL per mice each day.Sijunzi decoction and Danggui Buxue Decoction low dose group were reveived CTX 20 mg per kg weight each day and traditional Chinese medicine 0.1 g per mice each day.Sijunzi decoction and Danggui Buxue Decoction mediate dose group were reveived CTX 20 mg per kg weight each day and traditional Chinese medicine 0.2 g per mice each day.Sijunzi decoction and Danggui Buxue Decoction high dose group were reveived CTX 20 mg per kg weight each day and traditional Chinese medicine 0.4 g per mice each day.The weight indices of thymus gland and spleen,NK-cell activity and the proliferation of T-lymphocyte in five groups were measured after ten 10 days treatment.Results The weight indices of thymus gland and spleen,NK-cell activity and the proliferation of T- lymphocyte were increased in all three Sijunzi decoction and Danggui Buxue decoction groups,especially in Sijunzi decoction and Danggui Buxue decoction mediate and high dose group,the differences between them and CTX group were statistically significan(P<0.05,P<0.01).Conclusion Sijunzi decoction and Danggui Buxue decoction can antagonize the decrease of immunological function in tumor-bearing mice treated by chemotherapy.

ABSTRACT

[Objective] To investigate the influence of serum containing Danggui Buxue Decoction (DBD) on chemotaxis of oxidized low-density lipoproteins ( OxLDLs) for monocytes ( MC). [ Methods ] Six New Zealand rabbits were randomized to DBD group, Radix Angelicae Sinensis (RAS) group and Radix Astragali seu Hedysari (RASH) group. The rabbits were treated by gastric infusion of the above drugs respectively, bid for three days. On the third day, the serum containing drugs was obtained by heart blood sampling one hour after administration. Free chemotaxis group (normal control group) and OxLDLs group ( model group) were established for control. The number and figure of OxLDLs - chemotactic MC were observed by micropore filter method with chemotactic chamber. Object To establish the fingerprint of isoflavones and ferulic acid of Danggui Buxue decoction.Methods HPLC with Hypersil ODS column was adopted,the methanol-0.2 % acetic acid glacial(gradient elution) as mobile phase with a flow rate of 1mL/min and detecting wavelength at 254 nm.Results There were 10 main peaks in Danggui Buxie decoction,9 of them came from Radix astragali and 3 came from Radix angelicae sinensis.Conclusion This fingerprint can be used as a reference for the stability of the isoflavones and ferulic acid in Danggui Buxue decoction.

ABSTRACT

AIM: To investigate the protective mechanism of Danggui Buxue Decoction(Radix Astragali,Radix Angelieae Sinensis) on the kidney of diabetic rats. METHODS: The diabetic animal model was made by a single intraperitoneal injection of 60 mg/kg streptozotocin(STZ) and treated with Danggui Buxue Decoction(DBD) or Benazepril. Renal weight/body weight ratio, blood glucose and lipid, urinary albumin and ? 2-microglobin were measured. The renal pathological lesions were also investigated. The expression of TGF-? 1 mRNA in renal cortex as well as Angiotensin Ⅱ(AngⅡ) concentration of plasma and kidney tissue were determined by In situ hybridization and radioimmunoassay after 4,8 weeks of treatment. RESULTS:DBD had equivalent effects with Benazepril in protecting renal function and alleviating pathologic lesions. By means of inhibiting the expression of of TGF-? 1 mRNA. DBD had no effect on renal and plasmaAngⅡ. CONCLUSION:Danggui Buxue Decoction may alleviate kidney lesion and slow down the progression of DN by decreasing the renal expression of TGF-? 1 mRNA.
